NBC said Ramos was charged with five counts of first-degree murder.
Phil Davis, the Capital Gazette's crime and court reporter, began tweeting about the incident after he was safely away from the building.
The paper received threats via social media ahead of Thursday's shooting, officials said.
Prosecutors said Friday that the suspect barricaded the back door so that the victims could not get out and that one victim who tried to escape through a back door was shot.
